血行力学受容に伴う血管のチロシンリン酸化機構のリアルタイムイメージング解析

研究概要

平成12年度の研究においては、血管の組織・単離細胞での、機械的、物理的刺激受容から細胞機能発現に至るまでの情報伝達、いわゆるメカノトランスダクションにおいて、細胞膜近傍の接着因子タンパク系のSrcファミリーチロシンキナーゼ活性が、機械的情報を選択感受する機械センサー(メカノセンサー)であると仮定し、機械刺激とチロシンキナーゼ活性に関して以下の研究を行った。
1)メカノセンサーの過剰発現:酵素免疫法(ELISA)を独自に改良し、cSrcチロシンキナーゼ活性および阻害薬(herbimycin A 等)の作用を定量化した。cSrc527Lを過剰発現させたcos7細胞にストレッチ(150%伸展)や低浸透圧などの力学刺激を負荷すると、cSrcによってチロシンリン酸化をうける基質タンパクが有ることを見いだした。さらに、cSrcの発現の程度をGFP(green fluorescent protein)にて標識し、力学刺激の受容による細胞内Ca2+濃度の共焦点レーザー顕微鏡による同時測定を検討する。今後、血管平滑筋や内皮細胞を用いて同様の研究を計画している。
2)低浸透圧刺激とチロシンキナーゼ活性:低浸透圧と機械的伸展の力学刺激としての類似点と相違点をチロシンキナーゼ活性の観点から比較した。イヌ脳底動脈やその単離細胞標本では低浸透圧はその大部分が機械的伸展として作用することを見いだした。
3)低浸透圧刺激とイオン電流:低浸透圧刺激により増強するL型Ca2+チャネル電流に加えて、容量感受性Cl-チャネルや伸展活性化チャネル電流にたいする効果についてチロシンキナーゼ活性との関連について実験を行いつつある。
今後の計画:機械力学受容の細胞内情報伝達特異性、特にチロシンリン酸化がセリン・スレオニンキナーゼ活性(例えばプロテインキナーゼCやRho/Rhoキナーゼ)をどう調節し、血管機能を発現しうるかを可視化技術を駆使して研究する。
