イソプレノイド生合成におけるプレニル鎖延長酵素の触媒機能発現機構

研究概要

イソプレノイド化合物の生合成において最も基本的な過程である、C_5単位のプレニル鎖延長反応を触媒する酵素「プレニルトランスフェラーゼ」の内、シス型プレニル鎖延長酵素として初めて研究代表者らがクローン化した、Micrococcus luteus B-P26のウンデカプレニル二リン酸合成酵素(UPS)のX線構造解析を進め、2.2Åの分解能でホモ二量体構造の立体構造の解明に成功した。このUPSのコア構造は6本のβ-シートにより形成される、全く新しい折り畳み構造であった。興味深いことに、トランス型プレニル鎖延長酵素のファルネシル二リン酸合成酵素をはじめとする、イソプレノイド生合成関連の酵素の立体構造がα-ヘリックスのみによって構成される、"Isoprenoid Synthase Fold"と提唱されている共通の立体構造を持っているのに対し、このUPSはβ-構造の多い、新規な折り畳み構造であることが分かった。
部位特異的変異導入実験により、29D,33R,80R,77N,78Wなどの保存残基がアリル性基質の結合に、さらに197R,203R,216E,73F,74Sがホモアリル性基質IPPの結合に関与している保存性残基として同定され、これらの残基はいずれも触媒活性発現にも非常に重要であることが明らかとなった。
一方、トランス型プレニル鎖延長酵素の中で特異なグループである、中鎖プレニル鎖延長酵素(ヘキサプレニル二リン酸合成酵素およびヘプタプレニル二リン酸合成酵素)の解離性ヘテロサプユニットの各々について、反応生成物のプレニル鎖長の決定に関与するアミノ酸残基を追究した結果、大きいサブユニット側で反応生成物の鎖長決定に重要とされるDDXXDモチーフの上流のAlaやSerの他に、小サブユニット側に保存されている領域についても基質の結合や反応生成物のプレニル鎖長決定に関与する部位があることが判明した。
