分泌タンパク質の成熟化に伴う動態制御機構の解析

研究概要	1.小胞体で高次構造形成を終えた蛋白は、COPIIコートで被われた遷移領域から輸送されるが、その過程が様々な条件で阻害される事が知られている。そこで、そのCOPIIコートの細胞質プールとの交換反応について、生細胞においてFRAP(蛍光消光回復法)等を用いて調べると、多様なモードでの制御が起きている事を明らかにした。特に、浸透圧ストレスによる分泌阻害の際にはコートの離脱が特異的に阻害されていることが証明され、これはJNKの阻害剤で抑制されたのでCOPIIコートのGAP活性がJNKによる負の調節を受けている事を示唆した。更に、浸透圧ストレスと類似のコート離脱速度の低下が、cdckinase2の特異的な阻害剤によっても観察された。そこで、その阻害剤存在下における分泌蛋白の動態を調べると、foldした蛋白の小胞体からの輸送のうち、fast phaseは変わらないもののslow phaseの輸送が抑制されていた。このような事から構造形成を終えた蛋白の小胞体からの輸送機構において新たなモードが存在する事が示唆された。 2.fibrinogenは3つの異なる(しかし相似の)サブユニットが会合して分子間でジスルフィド結合を形成し、最終的に6個のユニットからなる会合体を作る。このうち、グルコースプロセシングが阻害されるとγ鎖の正しいタイミングでの会合体への組み込みが抑制される事を見いだした。 3.構造形成に失敗した分泌蛋白のプロテアソームによる分解をEDEMが促進する事を先に報告したが、これはERmannosidaseIの下流にあると考えられたので、過剰発現して分解への影響を調べたところ、EDEMとは異なる特性を示した。よって小胞体関連分解においてEDFM依存的な経路と非依存的准経路の2つが存在する事が示された。
