核内巨大酵素複合体による転写制御機構の解明

研究概要

わたしたちはCREB結合タンパク質(CBP)に代表される転写統合装置と呼ばれる分子群の解析をしている。特に、RNAヘリケースA(RHA)がRNAポリメラーゼII(pol II)をリクルートすることにより、CBPの転写活性化に必要であることを証明した(Cell 1996 ＆ 1997,G＆D 1997)。すなわち、アセチル基転移酵素(CBP)/ATPase・ヘリケース(RHA)/RNAポリメラーゼ(pol II)の酵素複合体が核内に存在することを発見した。本複合体の生物学的意義を検証することにより主として以下の成果が得られた。
1.RHAの最小活性化領域の決定(Aratani et al.,Mol.Cell.Biol.2001)
RHAの転写活性化領域(pol II結合領域)を生化学的に同定し、CREB依存的転写系での関与を検討した結果、その転写活性化領域が331-380番目の50アミノ酸から成り、中でも線虫からヒトまで完全に保存されているトリプトファン残基が非常に重要であることを見出した。
2.RHA複合体によるHIVウイルス活性化機構の分子機作(Fujii et al.,J.Biol.Chem.in press)
HIVの発現調節領域TARにRHAはそのRNA結合モチーフを介して結合することが明らかになった。また、同モチーフの236番のリジン残基をグルタミン酸に置換した変異体(K236E)ではTARに対する親和性が有意に低下していることも証明された。さらに、野生型RHAはHIVウイルス発現を転写レベルで活性化するが、K236E変異体、およびATPase活性の欠損した変異体ではこの作用が認められなかった。これらの結果をもとに、RHAをATPase/ヘリケース様因子としては最初のTARを介したHIV発現にかかわる宿主因子であることを報告した。
