研究概要

エピジェネティクスとは、遺伝情報を選択的に活用するために、DNAのメチル基修飾、DNA・RNAやタンパク質の分子間相互作用によって遺伝子が活性化または不活性化される巧妙な仕組みであり、転写、発生分化、発癌、複製、変異と修復、ゲノムの安定性などの多様な基本生命活動において重要である。近年、DNAメチル化酵素、メチル化CpG結合タンパク質、などの新規な分子・複合体が相次いで発見され、種々の基本生命活動が分子論的に解明されつつある。DNAメチル化は、これまで転写の不活性化に密接に関与することが示されていた。実際、発生過程ではDNAのメチル化が動的に変化して細胞の分化能が決定される一方で、多くの腫瘍では癌抑制遺伝子のプロモーターが高度にメチル化され、転写が抑制されていることが報告されている。近年、このメチル化されたDNAに結合し転写を抑制する因子として、Methyl-CpG binding protein 1(MeCP1)が得られた。MeCP1は、Methyl-CpG binding domain protein 2(MBD2)やヒストン脱アセチル化酵素からなる複合体である。MBD2には同一遺伝子から生じるMBD2a, MBD2bの二つのアイソフォームが存在しMBD2bはMBD2aに比べてN末端の152アミノ酸が欠失している。本研究にてMBD2a, MBD2bはともにメチル化されたCREの転写を抑制した。一方で、予想に反して、MBD2aのみがメチル化していないCREからの遺伝子発現を活性化した。結合実験により、MBD2aはCREB転写装置の構成因子であるRNAヘリケースA(RHA)と選択的に結合し、さらに、MBD2aとRHAは協調的にCREB依存的な転写を活性化した。また、RHAと結合しているMBD2aは、転写コリプッレサーであるHistone deacetylase 1とは複合体を形成していなかった。以上の結果から、MBD2aは転写抑制複合体と相互作用して転写を抑制するだけでなく、転写活性化複合体としても作用しうる可能性が示された。
