研究概要	1 in vitro再構成系の確立エンドサイトーシスの分子メカニズム解明のためには、エンドサイトーシスによる取り込み小胞の形成をin vitroで再現する実験系の確立が必要であると考えた。In vitroで人工脂質膜(リポソーム)を膜成分として脳細胞質と反応させることにより、大型(直径>1μm)のリポソームから直径100nm以下の小胞が多数形成される実験系を確立した。小胞形成にはATPとGTPが必要であった。さらに、形成された小胞の大きさ、数、相対的質量を動的光拡散測定装置を用いて測定することにより、小胞形成の定量化を可能とした。 2 in vitro実験系における膜脂質の機能、動態膜脂質が小胞形成に及ぼす影響を調べるために、膜脂質組成の異なるリポソームを上記のin vitro実験系に用いて、形成される小胞の量を比較した。その結果、phosphatidylinositol-4.5-bisphosphate(PIP_2)量の増加により小胞形成が促進されることを見い出した。そこでPIP_2に注目し、その動態を調べた。放射性ラベルしたPIP_2を含有するリポソームを用いて小胞形成させた後、PIP_2を測定した結果、小胞形成にともなってPIP_2はphosphatidylinositol-4-phosphate(PIP)、さらにphosphatidylinositol(PI)に分解されることが明らかとなった。 3 培養細胞のエンドサイトーシスによる膜脂質の動態上記in vitro実験系でみられた小胞形成にともなうPIP_2の分解が、細胞のエンドサイトーシスにおいて起こるか調べるため、PIP_2を代謝的に放射性ラベルした培養細胞(HeLa細胞)をエンドサイトーシス刺激した後、その動態を追跡した。その結果、同様にPIP2の分解がそれらの細胞においてみられた。
