糖ペプチド医薬の分子設計

研究概要	セレクチンと相互作用するシアリルルイスxとインテグリンβ1と相互作用するArg-Gly-Asp-Ser(RGDS)配列の両方を分子内に有するH-Lys[NeuAcα2-3Galβ1-4(Fucα1-3)GlcNAcβ-0-(CH_2)_3CO]-Gly-Arg-Gly-Asp-Ser-OH(糖ペプチド1)を合成化学的手法および酵素化学的手法をうまく連携させて効率よく合成することができた。糖ペプチド1はP-セレクチンあるいはE-セレクチンに対してシアイルルイスxよりも高い親和性を有しており、ヒトインテグリンβ1とそのモノクローナル抗体との結合をArg-Gly-Asp-Serよりも強く阻害することがわかった。また、表面プラズモン共鳴による分析では、このヘテロバイファンクショナルな糖ペプチド1はP-セレクチンおよびインテグリンβ1の両方と同時に安定な複合体を形成することができることもわかった。さらに、糖ペプチド1はインテグリンβ1を介した活性化ヘルパーT細胞のコラーゲンコートしたプレート上への接着も強く阻害した。これらの結果は、異なる生理機能を有する糖質とペプチドとを合成化学的手法および酵素化学的手法をうまく利用してハイブリッドさせることは、抗炎症剤や抗ガン剤の設計の新しいコンセプトとして非常に有用であることを示している。
