ポリアミノ酸の二次構造を利用する新規核酸吸着剤の開発

研究概要	本年度は、前年度までの基礎実験結果をもとに、試作吸着剤を製造し、その性能を評価した。またワクチン製造業者、試薬会社及び種々の研究期間へも試作吸着剤配付し、性能評価を依頼した。 1)試作吸着剤の製造:クロマトグラフィー法に使用するための試作吸着剤の基材としてセルロースビーズを製造した。製造は、チッソ株式会杜水俣工場で、従来のセルロース粒子製造法により製造した。粒径は、タンパク質等の有効成分を目詰まりなく通液させるために40〜100μmに設計した。また、核酸の選択吸着には粒子の細孔径が重要であるので、細孔径が糖の最大排除分子量(M_<lim>)で1×10^6と1×10^3の2種類の粒子を製造した。さらに同粒子にポリ(ε-リジン)(PL)を固定化し、試作用の核酸吸着剤(Cell-PL)を製造した。 2)核酸吸着システムの開発:末端ユーザーが試作吸着剤を用いて、エンドトキシンを簡便に除去できるように設計された吸着システムを開発した。 <バッチ法>小規模で、3時間程度で、簡便に行えるエンドトキシン吸着処理システムを開発した。 <カラム法>小規模(カラムサイズとして1mL-50mLを想定)で、研究者や医師がその場で薬液や培地をシリンジなどで通液することでエンドトキシンを除去できるプレパックカラムの開発を検討中である。 3)試作品性能評価上記試作吸着剤をパックしたカラムを用いて、タンパク質と核酸の混合溶液からの核酸の選択吸着除去を試みた。大きい細孔径(M_<lim>×10^6)をもつCell-PL-10^6吸着剤を用いた場合は吸着処理時の緩衝液の条件がpH7.0,イオン強度μ=0.05〜0.4で、小さい細孔径をもつCell-PL-10^3吸着剤の場合は、pH7.0,イオン強度μ=0.4〜0.8で、タンパク質(アルブミン)を吸着することなく、核酸を選択吸着できることがわかった。以上の結果より、今回調製したポリアミノ酸固定化高分子微粒子は、ワクチンや血液製剤等の注射溶液から、それらの有効成分にダメージを与えることなく、核酸のみを除去できることが明らかとなった。
