細胞表層工学技術による経口投与型の新しい抗原エピトープ提示生細胞ワクチンの開発

研究概要	酵母で働く分泌シグナルをN末端に、細胞膜にリン脂質を介して留まるGPI-アンカーシグナルをC末端にそれぞれ配置した、我々のこれまで開発してきた遺伝子工学的細胞表層提示システム、「細胞表層工学」、を用いて、多くの感染ウイルスのコートタンパクのエピトープ領域に相当する抗原ペプチドを、遺伝子工学的に経口でもまた注射としても人体にとっては安全な酵母細胞表面に輸送局在提示した酵母細胞を作製し、本抗原提示細胞をそのまま用いた一種のアジュバンド(免疫増強担体)一体化細胞ワクチンとして、特にヒトや家畜や養殖魚などへの新しい形態の生細胞ワクチンとしての活用をめざしている。昨年度は、抗原決定部位(エピトープ)程度のサイズの機能性ペプチドをコードする遺伝子を作製し、酵母の細胞表層で発現させたペプチドが活性を持つことを証明した。今年度は、提示産物が免疫原性を確保できるかを確認するために、提示したペプチドや分子サイズの大きな酵素タンパク質の細胞表層における提示量を蛍光分析と画像解析により定量した。この結果、直径5ミクロンの酵母表層に免疫原性をもつ提示タンパク質は約100万分子と算出され、十分な活性と抗原決定部位(エピトープ)としての力価を持ちうることが明らかになった。また、腸管免疫におけるマクロファージなどの攻撃サイズとしても、酵母は最適であることがわかった。これらの研究をふまえて、サケやマスなどの北方養殖魚に深刻な被害を与えているウイルスの抗原決定部位をコードする遺伝子を、我々のこれまで開発してきた遺伝子工学的細胞表層提示システムに導入し、新しい形態の生細胞ワクチンを作製し、実践応用への可能性を探った。
