黄体機能の調節によるウシの生殖制御技術の開発

研究概要

本研究は,着床・妊娠を通して中心的役割を持つウシ卵巣の黄体機能を制御して,最適な子宮環境を維持して受胎率を向上させる技術開発を目的としておこなった。特に黄体機能の局所調節機構に着目し,局所因子の制御によって血管新生や血管機能を刺激して強靭な黄体を作出する理論的な基盤とその具体的モデルを確立しようとした。本研究では,細胞間伝達機構の観察に適している微透析システム(MDS)を用いて,排卵卵胞と黄体機能の活性化および抑制の能力が高い局所生理活性物質(PG及び血管作動性ペプチド)の相互作用等を詳細に検討した。加えて,カラードップラー超音波画像診断装置を導入して,卵巣の個々の卵胞や黄体内の血流観察の方法を確立した。これらの独自に開発したウシ生体モデルを用いたことで,多くの卵巣内局所分泌と血流に関する新しい知見を得ることができた。さらに,ウシ黄体から単離した血管内皮細胞の培養系を用いて解析をおこなった。主な成果は以下の通りである。
1)排卵卵胞基底膜内のPGと血管作動性ペプチドが密接な相互作用によって排卵に至る血管機能と分泌機能を制御していることを見出した(排卵卵胞内のET-Ang-ANPシステムの提唱)。これらと,LHサージによる卵胞基底部の血流の急上昇が直接関係していることを発見した。
2)排卵後の黄体形成は,活発な血管新生に支えられているが,この機構にPGとAng-IIそしてANPが深く関わっていることを明らかにした。特に,この時期は黄体退行因子であるPGF2αがむしろ黄体刺激因子になっている事を提唱した。
3)成熟した黄体は,形成期と異なりPGF2αに対して反応するようになり,血管作動性ペプチドが血流の抑制に中心的な役割を果たしていることを,分泌と血流像から初めて示した。
4)排卵後hCG投与で黄体の血管新生を刺激することで強靭で大きな黄体作成の実証が進行中である。
