昆虫細胞による糖鎖エピトープ保持組換え型アレルゲンの生産と活性評価

研究概要

アスパラギン結合型糖鎖の付加サイトを持つ種々のタンパク質を昆虫細胞で発現させ、フコースを持つ糖鎖が付加された糖タンパク質を生産した。バキュロウィルスゲノムDNAに組み込むためのトランスファーベクターに各タンパク質cDNAを挿入した。ウィルスゲノムDNAと目的のcDNAを挿入したトランスファーベクターを昆虫細胞株、BTI TN 5B1-4に導入した。培養上清よりウィルスを回収し、β-ガラクトシダーゼ活性を指標にして組換え体ウィルスを選抜し、クローン化した。確立された昆虫細胞での発現系を用いて、2-3箇所のアスパラギン結合型糖鎖付加サイトを持つ数種の糖タンパク質については、大量に発現させ、分離・部分精製した。得られた組換え型タンパク質に糖鎖エピトープが付加されていることを、糖鎖エピトープ特異的抗体との反応性を酵素免疫測定法(ELISA)および免疫ブロット法を用いて解析した。その結果、組換えタンパク質の種類によって反応性に差がみられたものの、全て糖鎖エピトープ特異抗体との明確な反応が確認された。さらに、糖鎖構造のみを破壊する条件での過ヨウ素酸酸化処理により反応性が消失することを確認した。現在、さらに多種類の組換え糖タンパク質に生産を試みるとともに、糖鎖エピトープの構造を、種々のグリコシダーゼとレクチンを用いた解析、糖組成分析、およびアレルギー患者血清抗体との反応性の解析を進めている。本研究により、バキュロウィルスと昆虫細胞を用いた発現系で糖鎖エピトープを保持した組換え型糖タンパク質アレルゲンを生産でき、さらにその組換えアレルゲンが天然のものと同等のアレルゲン性を保持することが示唆された。このように昆虫細胞での糖タンパク質発現系はアレルギーに関する研究用試薬、診断試薬の生産系として有用であると考えられた。
