ペプチドスーパーアゴニスト同定法の開発とその応用

研究概要	コンビナトリアルペプチドライブラリーと質量分析を組み合わせることにより、T細胞が本来認識する野生型ペプチドよりもはるかに強い抗原活性を有するペプチドを効率よく同定する方法を開発し、これに改良を加えた。また、特異性未知の1個のT細胞を出発材料としてそのT細胞が認識する野生型ペプチドを同定することに成功した。具体的には、X19 random peptide, agonistic anti-CD29,IL-4,IL-7,L-9,IL-15を組み合わせることにより、末梢血から分離したばかりの新鮮なT細胞1個を、いきなりcloning conditionで培養し、10e7レベルに増殖させることができた。これらにより、自然界に存在し、なおかつ一次構造上似ても似つかないmimicry peptideを同定することも可能になった。また、擬態分子が誘導する免疫応答を考える際に、HLA class II分子を介して抗原提示細胞側に伝達されるシグナルも重要であることが明らかとなり、これに関しても成果をあげることができた。疾患との関連においては、ヒトT細胞抗原認識における分子擬態を免疫遺伝学的に解析し、環境要因としての抗原を新たな視点で捉え直すことを目的としている。これにより、感染症と自己免疫病との関連を分子レベルで明らかにすることを目指している。具体的には、環境要因(特定の感染症)と自己免疫病発症・増悪との関連を解明したい。
