遺伝性食後高脂血症を発症する機能遺伝子の探索

研究概要	1)食後高脂血症家兎の作出 WHHL家兎の中から常に高トリグリセリド血症を示す家兎を選抜・育種し、遺伝的に純系として確立した(TGHと名づけた)。TGHと日本白色家兎とからFl、F2世代を作出した所、F2世代にコレステロール値とTG値は絶食時に正常値を示していても、食後にだけTGの異常高値を示す個体を7羽(雄4羽、雌3羽)見出した。これら7羽を種動物としてF3世代を作出し、遺伝形式として垂直伝播性であること、劣性遺伝の形式をとる事を明らかにした。摂食開始後8時間目から高トリグリセリド血症が始まり24時間後に最大値に達した。食後高脂血症は4ヶ月齢から12ヶ月齢に顕著に観察された。成人型の食後高脂血症の病態に近似していることが明らかになった。 2)食後高トリグリセリド血症を来たす機能遺伝子の探索清潔開腹手術下に絶食時と食後18時間目に肝臓を生検した。これら2時点の肝臓サンプルからRNAを抽出してdifferential displayを行った。バンドの出現の差から機能遺伝子の発現の差を検討する事が可能となった。
