てんかん原性大脳皮質形成異常モデル動物の確立とアンチセンス治療法への展開

研究概要	1.focal freeze-lesionモデルの層構造形成異常の分子メカニズムの解析(岡部、福田): 生後0日齢ラットのfocal freeze-lesion法により局所的大脳皮質形成異常モデルを作成し、Cl^-トランスポーターのNKCC1(取り込み)とKCC2(排出)、および細胞移動関連分子reelinのmRNA発現状態をin situ hybridization法を用いて解析した。lesion部位における微小脳回の形成過程では、周囲の皮質板からのMAP2陽性の神経細胞の移動がlesion部位上部に認められ、この部位ではNKCC1 mRNA発現が増加していた。一方lesion内部ではGFAP陽性のグリア細胞が多数認められた。また、異常皮質形成後のI層でのreelin発現は正常部位と同様であった。 2.focal freeze-lesionモデルの層構造形成異常部位への移動細胞の電気生理学的解析(福田、井上): focal freeze-lesionモデルの障害部位に移動してくる神経細胞からスライスパッチクランプ法で膜電位記録を行った。これらの細胞を正常部位の錐体細胞と比較すると、電気生理学的膜特性がほぼ同様であるにもかかわらず、[Cl^-]_iが上昇しており、正常部位の細胞では起こらない、GABA/グリシンによる異常なCa^<2+>流入が認められた。周囲を電気刺激してもこれらの細胞でシナプス後電位は記録されなかった。以上は、未熟な移動中の皮質板細胞の特徴に類似していた。 3.ヒトてんかん原性皮質形成異常組織のCl^-トランスポーター・チャンネルとreelin発現の解析(清水、福田): ヒトのKCC2、NKCC1及びreelinのmRNAのオリゴcDNAプローブを合成し、外科手術により摘出された皮質形成異常組織を用いてin situ hybridization法を行った。ヒトてんかん原性皮質形成異常のひとつである皮質異形成組織では皮質は層構造を呈さずNKCC1の発現増加もなかった。異形成組織に特徴的な巨大細胞のKCC2発現は正常部位と同程度であったが、その周囲の小型細胞では発現が減少しており、異形成組織におけるCl^-調節の異常が示唆された。またreelin発現量は著しく低下していた。これらはfocal freeze-lesionモデル動物の所見(I-IV層のみの異常皮質を形成し、poly microgyriaの特徴に類似)とは明らかに異なっており、これらの成因が異なることが示唆された。
