カニクイザルを用いた胎児造血幹細胞遺伝子治療のモデル実験

研究概要	1.カニクイザル胎仔への移植 GFP(green fluorescent protein)遺伝子で標識した未分化ES細胞をカニクイザル胎仔4頭へ同種移植した。そのうち2頭は腹腔内へ、2頭は肝臓内へ移植した。腹腔内へ移植した2頭と肝臓内へ移植した1頭は移植1ヶ月後に、肝臓内へ移植した残りの1頭は移植3ヶ月後に、それぞれ帝王切開によって取り出した。移植1ヶ月後に取り出したサルでは、GFPを指標とするin situ PCRによって多数の胎仔組織に移植細胞を検出した。移植細胞の占める比率はreal time PCRで1%以下であった。テラトーマは認めなかった。移植3ヶ月後に取り出した胎仔では胸腔内にテラトーマを1個認めた。一方、多数の組織に、テラトーマを形成することなく移植細胞を認めた。移植細胞め古める比率はreal time PCRで1%前後であった。移植細胞は集蔟せず孤立して存在し、周囲の細胞から区別できない形態を呈していた。以上の結果から、カイニクイザル胎仔には、移植したES細胞を生着できるスペースがあって、このスペースにうまく「はまりこんだ」細胞は、周囲の微小環境から適切な増殖・分化のシグナルを得て生着し分化を遂げるが、胸腔内などに「もれた」細胞は周囲から適切なシグナルを得ることができずテラトーマを形成すると推察された。また、テラトーマ形成結果から、ES細胞移植応用にあたっては未分化ES細胞の除去の重要性が明らかである。 2.ヒツジ胎仔への移植血液細胞に分化誘導したカニクイザルES細胞の中に造血幹細胞が含まれるかどうかを調べるために、これらの細胞をヒツジ胎仔へ異種移植した。これらの胎仔のうち3頭はすでに自然分娩によって生まれた。3頭のうち2頭のヒツジ体内にサルの造血細胞が出来ていることを確認した。今後は、ヒツジの体内においてサルの造血が占める比率を上げる工夫を行う一方、造血幹細胞へ分化誘導したカニクイザルES細胞をカニクイザル胎仔へ同種移植する実験を予定している。
