PPARγ内因性リガンド同定による肥満・インスリン抵抗性の新規診断・治療法の開発

研究概要	合成のPPARγアゴニストによるPPARγの著明な活性化は、脂肪細胞分化を誘導して、白色脂肪組織での脂肪蓄積を強力に促進し肥満を助長するが、そのことによって骨格筋や肝臓における中性脂肪の蓄積を抑制し、インスリン抵抗性を改善させている。また脂肪細胞分化によって新たに生じた小型脂肪細胞の増加とアポトーシスによる大型細胞の減少により、脂肪細胞を小型化させることによって、インスリン抵抗性惹起分子である遊離脂肪酸やレジスチンを低下させ、インスリン感受性ホルモンであるアディポネクチン(Nature Medicine 7 : 941-946, 2001)を増加させて、インスリン抵抗性を改善させていることを報告した(J.Biol.Chem.276 : 41245-41254, 2001)。さらに欠損マウスを用いてCBPがPPARγの主要なコアクチベー夕ーであり、PPARγによる脂肪細胞サイズ、インスリン感受性調節に重要な役割を果たしていることを示した(Nature Genetics 30 : 221-226, 2002)。また脂肪細胞分化にはIRS-1/IRS-2がPPARγの発現誘導を介して重要な役割を果たしていることを示した(Mol.Cell.Biol.2 : 2521-2531, 2001)。我々は脂肪細胞分化に伴ってインスリン感受性を獲得する3T3L1脂肪細胞よりPPARγ内因性リガンドの精製を試みた。PPARγの活性化能を指標にHPLC上1本のピークに精製した。そのピークのretention timeは既報のマクロファージの内因性リガンドの第一候補である15d-PGJ2や脂肪酸代謝産物と異なっていた。また、そのPPARγ活性化能はHPLC上認められるピークの高さからすると、合成リガンドとしては最も活性化能の高いロジグリタゾンと同程度かそれ以上と考えられた。質量分析計上10程度の候補分子が存在したので、精製材料をin vitroのものからin vivoのものへ変更し、現在候補分子を絞り込んでいる。
