多因子疾患における機能ゲノム学の構築:糖尿病をモデルケースとして

研究概要	ポストシークエンス時代のゲノム医学の大きな課題は遺伝子変異と疾患の機能的関連を解明すること(機能遺伝学)である。特に、糖尿病をはじめとする多因子疾患において、遺伝子間の相互作用ならびに遺伝子と環境因子の相互作用を解明し、複数存在する疾患感受性遺伝子が個々にどのような機能を介して疾患発症に寄与するかの全貌を解明し、適切な予知法・予防法・治療法の構築につなげることが不可欠である。遺伝的背景の不均一な人においてこれを達成することは不可能であることから、我々は近交系モデル動物を用いて遺伝的背景ならびに疾患を均一にするとともに、その一部を改変したコンソミック系統・コンジェニック系統を作出することで個々の遺伝子機能を単独で解析した。対象としてヒト2型糖尿病の優れたモデル動物であるNSYマウスを用いた。 NSYマウスに蔗糖負荷、高脂肪食負荷を行ったところ、いずれにおいても著明な体重増加をきたしたが、蔗糖負荷においてのみインスリン分泌低下を伴う糖尿病早期発症を認めた。NSYマウスを対象に施行したゲノムスキャンの結果、マップすることに成功した3つの主遺伝子(Nidd1,Nidd2,Nidd3)をそれぞれ単独で有するコンソミックマウスを作出した結果、Nidd1はインスリン分泌障害と抵抗性を基盤に血糖上昇に関与、Nidd2はインスリン抵抗性のみを基盤に血糖上昇に関与、Nidd3は脂肪分布・脂肪肝・インスリン抵抗性に関与することが明らかとなった。さらに、Nidd1とNidd2の両者を有するダブルコンソミックマウスを作出し、機能解析を行った結果、単独のコンソミックに比し、2つの遺伝子の共存により著明な血糖上昇を認めたことから、Nidd1とNidd2の遺伝子間相互作用により糖尿病の表現型が増強されることが明らかとなった。
