脳腫瘍特異抗原遺伝子のクローニングと免疫遺伝子治療への応用

研究概要	悪性脳腫瘍は極めて治療困難な疾患であり、現在まで有効な治療法がない。そこで申請者らは新たなアプローチとして、脳腫瘍に対する腫瘍抗原特異的な免疫遺伝子治療を提案している。本研究では第一に新規の脳腫瘍特異抗原遺伝子の単離を目的として、グリオーマ患者血清を用いてautologous typingに分子生物学的手法を取り入れたSEREX(Serological identification of antigens by recombinant expression cloning)法による解析を試みた。患者一症例につき5x10^5個のファージを12例のグリオーマ患者血清を用いてスクリーニングした結果、92個の陽性クローンを単離し、塩基配列解析により、それらの抗原は52種の遺伝子をコードしていることが明らかになった。さらに単離された抗原を、様々な患者(グリオーマ患者、他の癌患者、他の脳疾患患者、および健常人)血清中IgGとの反応を解析し、グリオーマを含む癌患者のみ反応する新規遺伝子KU-GB-1の全長を単離した。一方、癌ワクチンに用いるベクターとして、in situで遺伝子導入効率が高く、腫瘍細胞中でのみ選択的に複製するreplication-conditional HSV、G207をヘルパーウイルスとして用いて、サイトカイン遺伝子(GM-CSF及びIL-12)をamplicon vector組み込んだdefective HSV vectorシステムを作製した。
