免疫担当細胞および免疫抑制剤による神経栄養因子産生を介する脊髄損傷修復の基礎研究

研究概要	免疫制御剤には本来の免疫抑制作用のほかに神経栄養効果が報告されてきた。本研究によってシクロスポリンAやタクロリムスが脳由来神経栄養因子(BDNF)やグリア細胞株由来神経栄養因子(GDNF)の産生を促進しこれを介して神経栄養を発揮することを明らかにした。しかし神経保護作用を期待するとき免疫抑制作用は生体に不利益をもたらす恐れがある。そこで本年度は免疫抑制作用をもたないイムノフィリンリガンドリガンドの探索と薬効判定を試みた。タクロリムスがイムノフィリン(FKBP12)に結合する構造に注目し、これと類似した構造を持つLeu-Ileジペプチドとその類縁物質「A」について検討した。両物質は神経栄養因子の産生を促進し、培養下で神経細胞死を抑制すること、さらに培養脾臓細胞によるインターロイキン2の産生を抑制しないことを明らかにした。以上の結果よりいずれの物質も免疫抑制作用をもたない新しいイムノフィリンリガンドであると考えられたので、胸髄を半切断した脊髄損傷ラットを作製し、「A」物質を腹腔内に毎日投与した。投与開始3日目から歩行能力が回復し23日後にはほぼ損傷前のレベルまで運動機能が回復した。無処理の脊髄損傷群も時間の経過に伴い緩やかに運動機能が上昇したが投与群より低いレベルにとどまり、16日後以降の回復は観察されなかった。以上、[A]物質は脊髄損傷の修復に有効であることが示唆された。Leu-Ileジペプチドについても現在検討中である。これらの免疫抑制作用をもたないイムノフィリンリガンドをリード化合物してさらに強力な活性物質が創製できれば、脊髄損傷をはじめとする中枢神経疾患の治療薬の開発につながるものと期待される。
