軟骨誘導遺伝子を利用した自家細胞移植による軟骨/骨の再生

研究概要

軟骨分化における軟骨由来の転写因子であるDEC1の発現パターンを検討した。その結果、ATDC5細胞株では、インスリン処置によって軟骨細胞へと分化するにともなって、DEC1mRNAレベルが100倍上昇することが判明した。またウサギ骨髄由来間葉系細胞の培養系で、DEC1mRNAレベルが軟骨型proteoglycan(aggrecan)の発現に先立って上昇することが判明した。ウサギ骨髄由来間葉系細胞の培養系でMTf遺伝子は、DEC1mRNAレベルの上昇より遅れて、aggrecanの発現に平行して上昇した。また予備実験において、ビーグル犬の歯槽骨欠損モデルに骨髄由来間葉系細胞を移植すると、歯槽骨が再生することが観察された。したがって、DEC1およびMTf遺伝子は骨髄由来間葉系細胞による軟骨/骨再生に関与していることが示唆された。またDEC1を条件的に発現するウイルスベクターを作成して、現在そのテトラサイクリン応答性を検討している。
