軟骨誘導遺伝子を利用した自家細胞移植による軟骨/骨の再生

研究概要	マウス胚性癌細胞由来ATDC5細胞株およびウサギ骨髄由来間葉系幹細胞に私たちが発見した転写因子DEC1を導入することにより軟骨分化を誘導することができた。DEC1は軟骨初期分化マーカーであるaggrecan, type II collageの遺伝子発現を誘導したのみならず、最終分化マーカーであるtype X collage, alkaline phosphatase, Runx2遺伝子の発現も強力に促進して、石灰化も誘導した。軟骨分化を促進する、BMP, TGF-b, insulinはDEC1の発現を促進するので、これらの成長因子の作用をこの転写因子が仲介していることが明らかになった。この知見はJ.Biol.Chem.2002に発表した。またDEC2もDEC1と同様に軟骨分化を誘導した。さらに膜結合型トランスフェリン様蛋白(MTF)が上記の成長因子によって誘導されること、そしてMTFが軟骨分化を促進することを明らかにした。この知見もJ.Biol.Chem.2002に報告した。軟骨分化は低酸素下で進行することから、低酸素がDEC1とDEC2の発現に及ぼす影響を検討したところ、DEC1とDEC2はHIF1を介して低酸素によって誘導されることも判明した。これもJ.Biol.Chem.2002に報告した。以上のように、軟骨誘導遺伝子を利用して骨軟骨疾患を治療する枠組みを構築することができた。
