研究概要

抗体は生体成分の微量分析に不可欠であるが、低分子化合物に対する抗ハプテン抗体の親和力には限界があるため、attomoleレベルの感度を得ることは不可能に等しい。そこで、従来調製が困難とされる抗メタタイプ(Met)抗体(抗原抗体複合体を特異的に認識する抗体)をキーマテリアルとするハプテンの高感度非競合型イムノアッセィの構築を企画した。抗Met抗体を調製するためには、抗ハプテン抗体とハプテンの複合体で動物を免疫することになるが、解離を防ぐために抗体のパラトープをハプテンでアフィニティーラベルすることが望ましい。そこで、昨年度はモデルハプテンとしてデオキシコール酸(DCA)を取り上げ、DCAアデニレートをラベル化試薬として用いるマウスモノクローナル抗DCA抗体(Mab)のアフィニティーラベル法を確立した。一方、抗ゼノタイプ抗体や抗アロタイプ抗体の副生を抑えるために、抗ハプテン抗体から定常部を除いた一本鎖Fvフラグメント(scFv)を免疫に用いるのが有利と考えられる。この観点から、本年度は上記MabのH鎖及びL鎖の可変部ドメイン(V_H、V_L)を連結したscFvの調製を検討した。抗DCA抗体産生ハイブリドーマからRNAを抽出し、5'RACE法によりV_H及びV_Lの遺伝子をクローニングし、塩基配列を決定した。また本データから、相補性決定部(CDR)のアミノ酸配列を特定した。ついで、V_H、V_Lの両遺伝子をoverlap extension PCRによりリンカー配列を介して連結し、scFv遺伝子[5'-V_H-(gly_4ser)_3-V_L-3']を構築した。これを大腸菌XLOLR株中で発現させて可溶型のscFvを調製したところ、由来するMAbの親和力と特異性を十分に保持していることが判明した。本scFvをDCAアデニレートでラベル化したのち免疫投与することで、目的の抗Met抗体が効率良く産生されるものと期待される。また、CDRの遺伝子にランダム変異を加えた変異scFvのライブラリーを構築することにより、動物が産生する天然の抗体を凌ぐ結合特性を有する"超抗体"の探索が可能と思われる。
