質量分析による糖蛋白質糖鎖高感度構造解析法の開発

研究概要	二次元電気泳動などから得られるピコモル以下の微量糖蛋白質糖鎖の構造解析を確立する目的で、1)電気泳動ゲルから分離された糖蛋白質を効率よく抽出、回収する方法、2)糖鎖切断の後に糖鎖を蛋白質部分と分離する方法、3)得られた遊離の微量糖鎖に対して行う4-アミノ安息香酸2-ジエチルアミノエチルによる化学誘導化法、4)高感度質量分析のための試料前処理法等の検討を行った。その結果、糖蛋白質のペプチド・マス・フィンガープリント(蛋白質データベース検索に利用)については、500fmol量から得られたゲルバンドに対して、インゲルーアルキル化、酵素消化、糖鎖切断酵素(PNGase F)による処理、逆相担体による脱塩処理を行うことができ、従来より多くの情報量を質量分析から得ることが可能となった。一方、逆相担体による脱塩処理で素通り画分に得られた遊離の糖鎖については、化学誘導化におけるロス、及び、低分子量の水溶性の共雑物のため、現在のところ、50pmol量の糖蛋白質から得られたゲルバンドに対して質量分析による検出、構造解析が可能であった。また、誘導化糖のタンデム質量分析において観測された断片イオンを系統化し、糖鎖の配列情報を簡便に得る方法(ソフトウェアを含む)を確立した。当初試作予定のミクロリアクターは糖鎖をゲルから単離した後に行う化学誘導化を効率よく行うためのものとして考えたが、上記結果より、最終的に得られる誘導化糖鎖の大幅な収率の向上にはつながらないと判断し、試作を断念した。次年度は、ゲル中の糖鎖を直接化学誘導化し、その後に逆相担体による単離、精製を行う方法を試み、より微量の糖鎖の検出、構造解析法を確立する。
