研究概要

研究代表者の大石、研究分担者の吉嶺がウガンダに出張し、Joint Clinical Research Centre(JCRC)において日和見感染のないHIV感染者(40名)と健康なHIV非感染者(20名)を半年間かけて登録した。HIV感染者は末梢血を用いたフローサイトメトリー法で末梢血CD4+Tリンパ球数(CD4)>200/μl(20名)とCD4<200/μl(20名)の2群に分けた。また、HIV感染者では血漿中のHIV-1 RNAを定量した。これらの60名のHIV感染者と非感染者の末梢血単核球を分離し、in vitroでlipopolysaccaride(LPS)+/-の条件で3時間培養し、RNAサンプルを抽出し、
東京大学医学部衛生学教室へ送付した。RNAサンプルはT7RNA transcriptaseで増幅し、これらを逆転写酵素反応によりcDNAとして、蛍光色素にはCy3,Cy5を用いてcDNAを蛍光標識した。
DNA arrayではT細胞、単球、樹状細胞などに発現し、情報伝達や、分化、宿主感染防御に作用する計500種の遺伝子をPCR法で増幅し、probeを貼り付けるためにポリL-リシンで加工したスライドガラスにスポッティングした。このoligonucleotide probeに蛍光標識したcDNAをhybridizationし、各々のprobeからのシグナルをスキャナーで検出し、コンピューター解析のシステムを確立した。現在、このシステムを使ってRNAサンプルの解析を行っている。
