研究概要

RS447メガサテライトDNAはヒト4p15に20〜100コピータンデム反復し多型と変動に富む。RS447単位長4746bpに1590bpのORFがあり脱ユビキチン化酵素USP17をコードし、少なくとも哺乳動物種によく保存されていた。以上より本研究では、ゲノム進化研究と実験解析のためのモデル系開発も視野に入れ、単離していたチャイニーズハムスター、ラットのRS447相同配列の解析とヒトRS447導入マウスの系統化を試みた。データベースのマウスDUB-1、-2配列も加え比較したところ、ヒトとの相同性はどの種もほぼ60%と同等である。反復数はチャイニーズハムスター75コピー、ラット10コピー、マウスではともに1コピーである。99.3%以上の種内相同性がありながら種間ではマウス-ラット間の77%が最も高く協調進化の可能性をすでに指摘した。ヒトでは第4染色体にほとんど並び8番にも数コピーある。ラットは中期染色体FISHとマッピングから第1染色体のみにタンデムに存在した。マウスDUB-1,-2はともに第7染色体と報告されている。マウス7番とラット1番はシンテニー領域があるがヒト4pには知られていないので新しい発見の可能性が高い。ヒトRS447をタンデム導入したトランスジェニックマウスではトランスジーンの遺伝性が低くホモ接合体は未だ得られていない。タンデム構造のRS447配列がマウスに何からの有害な作用をしていることを示唆する。ヒトではアンチセンス側の転写が認められ、とくに脳において顕著であった。ラットでは脾臓で強い転写が確認できたのみでアンチセンスを含めヒトにおける発現と異なっている。authologかparalogかの確認が必要となった。今後、体細胞レベルで高頻度変動を示すマウスp^<un>変異の解析などとも合わせゲノム変動と進化の関係をさらに解明する系となることが期待される。
