ペルオキシソームタンパク質の細胞内輸送を担う新規の因子の同定とその機能解析

研究概要	アラビドプシスped2突然変異体は、ペルオキシソームの機能の一つである脂肪酸β酸化系を欠損する。申請者は、この突然変異体を用いることによってペルオキシソームタンパク質の細胞内輸送機構の分子機作が解明できると考えている。本年度は、(1)ped2突然変異体のペルオキシソーム機能、(2)各種ペルオキシソームタンパク質の細胞内輸送状況、(3)PED2遺伝子およびcDNAの同定、(4)遺伝子産物の解析、の4点について検討を加えた。その結果、以下のことが明らかになった。 (1)ped2突然変異体は、脂肪酸β酸化系のみならず、光呼吸活性など、広くペルオキシソーム機能を欠損する。 (2)ペルオキシソームタンパク質には、細胞内輸送機構の異なる2種類のタイプが知られている。ped2突然変異体では、どちらのタイプに属するタンパク質も正しくペルオキシソームへ輸送されず、サイトゾルに蓄積している。 (3)分子遺伝学的解析の結果、PED2遺伝子がP1クローン;MQB2の7.2kb Xho I断片に存在することが判明した。cDNAの同定を行ったところ、この遺伝子には12個のエクソンが存在し、507アミノ酸からなるポリペプチドをコードすると推定された。アミノ酸配列は、ヒトPexl4pと約30%の類似性を持っており、本タンパク質をAtPexl4pと命名した。 (4)AtPexl4pは、75kDのペルオキシソーム膜タンパク質であり、その大半がサイトゾル側を向いている。以上の結果は、EMBO J.(2000)19,5701-5710.にて発表した。
