頭足類マダコの循環および生殖系に作用する神経ペプチドの生理・生化学的研究

研究概要

マダコ脳から新奇のバソプレシン/オキシトシン超族ペプチドを単離し、オクトシンと名付けた。大静脈神経内分泌系から既に同族のセファロトシンが単離されている。ほとんどの脊椎動物にはバソプレシン族とオキシトシン族のそれぞれのペプチドが存在するが、無脊椎動物では、一種の動物から単一種のペプチドしか見つかっておらず、単の分子種が、浸透圧調節と生殖器をターゲットとする二つの機能を合わせ持っていると考えられてきた。マダコで初めて二種のペプチドが発見されたことになる。それぞれの前駆体遺伝子をクローニングし、構造を比較すると、シグナル領域に続いてペプチドをコードする領域と、バソプレシン/オキシトシン超族ペプチド前駆体に特徴的なニューロフィジンドメインを持つことが分かった。それぞれのニューロフィジンドメインが全く同じ遺伝子でコードされていた。このことは、両者の遺伝子が、スプライシングバリアント、もしくは遺伝子重複によって生成した可能性がある。また、それぞれの前駆体遺伝子の異なった領域をプローブとして、脳における分布をin situ hybridization法によって調べた。その結果、オクトシンは、食道上脳の知覚や記憶、随意運動などを司る領域に主に発現し、一方、,セファロトシンは、食道下脳の内臓機能など不随意運動を司る領域と大静脈神経内分泌系に主に発現していることが分かった。
