組換えα-マンノシダーゼの構造と活性に関する研究

研究概要

本研究の対象とする1,2-α-マンノシダーゼはタンパク質のアスパラギン残基に結合しているN-アセチルグルコサミン2残基を介して結合しているハイマンノースタイプの糖鎖のうち、1,2-α-マンノシド結合のみを特異的に加水分解する酵素である。本研究では、すでに本酵素の詳細な酵素化学的研究を終え、さらに本酵素をコードする遺伝子msdSをクローニングし、酵母、ならびにコウジキンにより遺伝子産物の大量発現に成功している。
本研究に用いているクロコウジキンAspergillus saitoiの1,2-α-マンノシダーゼにはカルシウムの関与は全く知られていなかった。しかし、今回本酵素分子1モル当り1グラム原子のCa^<2+>の存在が知られた。このCa^<2+>は非常に固く結合していて、キレイト試薬であるエチレンジアミン4酢酸(EDTA)の処理によってもなかなかはずれなかったのだが、弱アルカリ性で処理することにより活性を失ったCa^<2+>フリーのアポ酵素が得られた。アポ酵素の熱分析の結果は大幅に変性温度を低下させた。弱アルカリ性のpH処理のみでは、円二色性(CD)の変化はほとんど無いのだが、弱アルカリ性のpH処理をしたCa^<2+>フリーのアポ酵素は円二色性(CD)の変化がみられた。以上の結果、クロコウジキンAspergillus saitoiの1,2-α-マンノシダーゼに活性に必須のCa^<2+>の存在を明らかにした。さらに、本酵素の活性はパラクロロメルクリ安息香酸により全く変化しないことから、コウジキンのα-アミラーゼのに見られるようなシステイン残基に結合したCa^<2+>ではないことが結論された。
