赤潮プランクトン、シャットネラの活性酸素産生を司る酵素系の解析

研究概要

Chattonellaのスーパーオキサイド産生がレクチンの一種であるコンカナバリンA(ConA)、あるいは糖特異性が異なる他のレクチン(リシン、CBH、WGA等)添加によって、著しく上昇すること、さらに興味あることに、魚鰓粘液由来高分子物質にも同様な刺激作用があることを見出している。これらの結果からChattonellaにも、哺乳類白血球同様、細胞外からの刺激に応答してスーパーオキサイド産生量を増加させる刺激応答機構あるいは酵素系(NADPH oxidase)が存在すると考えられる。さらに、Chattonellaの粗抽出液中にNADPHを添加すると、スーパーオキサイドが酵素的に産生されることを見出した。そこで、本年度はChattonellaの活性酸素産生に関与する酵素系、特に白血球のNADPH oxidaseとの類似性について検討した。まず、Chattonella細胞を超音波処理あるいは単に軽度の撹拌処理した後、細胞本体を遠心分離により除去し、上清を得た。これまでの予備実験により、この様な処理によりChattonella細胞表層構造であるグリコキャリックスが細胞本体から脱落することをを見出している。この上清にNADPHを添加するとスーパーオキサイドが酵素的に産生されることが確認されたので、これを粗酵素としてSDS-ポリアクリルアミドゲル電気泳動によりその構成成分をさらに細かく分離後、常法に従い免疫抗体法によりウエスタンブロットによる解析を行った。すなわち、ヒト白血球由来NADPH oxidaseと免疫学的に類似した成分が存在するか調べた。その結果、ヒト好中球のNADPH oxidaseの主要成分蛋白質であるチトクロームb大サブユニットに対する抗体と反応する主要バンド(110kD)が検出された。以上の実験結果により、Chattonellaのスーパーオキサイド産生を司る酵素系は哺乳類白血球細胞膜酵素であるNADPH oxidaseに類似していることが強く示唆された。
