鳥類の脳内浸透圧受容体と下垂体神経葉ホルモンの分泌調節機構

研究概要

哺乳類において、アクアポリンは水チャンネルとして同定され、腎臓などにおいて機能の研究が進んでいる。このアクアポリンのサブタイプであるアクアポリン4(AQP4)は、脳内に存在し水分のセンサーとして調節に関与しているのではないかと考えられている。鳥類においては、アクアポリンの研究は進んでおらず何も報告がされていない。本研究では、ニワトリの脳のAQP4をクローニングし、その局在性を明らかにし、AQP4が浸透圧受容体であるかどうかを明らかにしようとするものである。
哺乳類などのクローニングされた塩基配列から、プライマーを設計する。そして、ニワトリの視床下部から抽出したRNAを逆転写しDNAにしたのち、哺乳類から決めたプライマーを用いてPCRを行い、PCRサンプルを電気泳動し一つのバンドが確認し、プラスミッドにサブクローンし、塩基配列を調べた。この塩基配列が、相同性などから哺乳類のAQP4と非常に相同性が高いものであることであることから、この遺伝子断片はニワトリのAQP4であることが示唆された。今後、AQP4であるならば3'-RACEあるいは5'-RACEの方法を用いてAQP4の塩基配列の全長を決定し、また浸透圧などの生理的条件を変化させた場合の遺伝子発現を調べる予定である。
