消化管平滑筋での抑制性伝達の細胞内機序

研究概要

ラット近位結腸縦走筋標本において、一酸化窒素がcyclic GMP-PKG系を介さずに、平滑筋細胞内のCa^<2+>濃度を低下させる機序を検討し以下の結果を得た。
(1)電位依存性Ca^<2+>channel関与の有無について、一酸化窒素による張力とCa^<2+>濃度の変化を、電位依存性Ca^<2+>channel阻害薬の張力とCa^<2+>濃度への影響とを比較検討した。また、電位依存性Ca^<2+>channel阻害薬存在下の一酸化窒素の弛緩作用を調べた。その結果、一酸化窒素の弛緩作用への、電位依存性Ca^<2+>channelの関与がないことを確認した。
(2)細胞膜Ca^<2+>-ATPaseの活性化によるCa^<2+>の細胞外への汲み出しと、小胞体のCa^<2+>-ATPase(SERCA2)の活性化による小胞体へのCa^<2+>の汲み入れ、への一酸化窒素の関与について、標本をThapsigargin処置後に一酸化窒素による弛緩反応を調べた。その結果、一酸化窒素の弛緩作用が消失したので、SERCA2関与の可能性が肯定された。
(3)一方、スキンド標本でCa^<2+>濃度が一定であるのに、実際にcyclic GMPが弛緩を生じることを既に確認している。従ってこの弛緩の経路と先のcyclic GMPを介さない経路との関連を、PKG阻害薬のRp8-bromo cGMPSを用いて調べた結果、PKGを介するCa^<2+>感受性低下機構の生理的意義は薄いと考えられた。
