アドヘレンスジャンクションにおける新規細胞接着機構ネクチン・アファジンの役割

研究概要

新規細胞接着機構ネクチン・アファジン系の局在と機能の解析
これまで私共は、ネクチン・アファジンが小腸上皮細胞のアドヘレンスジャンクションに局在することを示した。そこで次に、従来からアドヘレンスジャンクション様構造を持つと報告されてきた脳のPuncta adhaerentia junctionなどの構造にもネクチンとアファジンが存在するか否かを光顕・電顕レベルで解析した。その結果、アファジンには大小2つのアイソフォームが存在し、脳では、大アファジンは海馬のCA3領域のmossy fiberの神経終末にのみ限局して発現しているのに対して、小アファジンは脳全体に広く発現していた。またネクチンにも1型、2型、3型の3種のアイソフォームがあり、それぞれのアイソフォームがどのアドヘレンスジャンクション様構造に局在するかを解析した。その結果、ネクチン1型がmossy fiberの神経終末に、ネクチン3型がCA3領域錐体細胞樹状突起に局在することがわかった。さらに、ネクチン1型機能を阻害する物質、gD.を海馬培養細胞に作用させると、シナプスの直径が減少し、個数が増加することがわかった。このことから新規細胞接着機構ネクチン・アファジン系は、シナプス形成に重要な役割を果たしていることが示唆された。
