GCP症候群の相同疾患マウスにおける責任遺伝子Gli3とShh遺伝子の発現異常

研究概要	Pdn/Pdnマウスでは、責任遺伝子であるGliの極端な発現抑制が明らかになっていたので、Gli3遺伝子の突然変異部位の特定に努めてきた。その間に、Rutherらによって、Pdnマウスでは、Gli3遺伝子のイントロンにトランスポゾンが挿入されていることが報告され、そのトランスポゾンがイントロン3に挿入されていることを明らかにしてきた。本研究において、このトランスポゾンの正確な挿入箇所と実態の解明を目指した。エクソン3と4にプライマーを設計し、LA-PCRを行った結果、約11KbのPCR産物が得られ、これをシークエンスした。その結果、エクソン4側の約10Kbが解読された。その後、エクソン3の下流1.8Kbの配列を明らかにした。結果的にはイントロン3は65.8Kbもあり、そこで、これらの塩基配列をNCBI Annotation ProjectのデータベースでBLAST検索した結果、Intron3はNW_000072に含まれることが判明した。また、Gli3遺伝子のイントロン3の上流から41286/41287bpに5442bpのトランスポゾーンが挿入されていることが判明した。 Pdn/Pdn,Pdn/+,+/+胎仔の遺伝子型を胎生早期より識別する簡便な方法を開発するために、トランスポゾン挿入部位の上流にプライマーF5を、下流にR6を、トランスポゾンにetn11Fを設計した。その結果、+/+では214bp(F5⇔R6)だけの、Pdn/+では214bp(F5⇔R6)と180bp(etn11F⇔R6)の、Pdn/Pdnでは180bp(etn11F⇔R6)だけのバンドが得られた。Pdnマウス胚の卵黄嚢膜からgenomic DNAを抽出し、この方法で遺伝子型を決定後、胚本体からRNAを抽出し、競合RT-PCRを行い、Gli3とShhの遺伝子発現量を測定した。Gli3遺伝子発現量は、いずれの胎齢でも、Pdn/Pdn、Pdn/+は、+/+に比べ発現抑制されていたが、Shhでは胎生9日に限って、Pdn/PdnやPdn/+は、+/+より高い値を示した。
