代謝型受容体刺激により活性化されるCa2+流入チャネルの分子生理学的解析

研究概要	形質膜越えのCa流入は、細胞内Caイオン濃度の主要な調節機構の一つであると同時に、膜電位の脱分極を担うという生理的意義を有する。最近、Ca流入経路として、受容体介在型陽イオンチャネル(Receptor-mediated cation channel)(RMCC)群が注目を集めている。TRPホモログ(TRP1-7)を見い出し、これらのTRPの組み換え発現により得た、単一分子種よりなる受容体活性化Caチャネル(RACC)の機能的解析を行い、"native"RACCの実体を探った。classicalなTRPホモログに加え、数10microMのH_2O_2等、活性酸素種によって活性化開口する新規Ca透過型カチオンチャネルLTRPC2を同定した。本LTRPC2チャネルの直接の活性化トリガーは、ニコチンアミド酸化体と考えられる。また、活性酸素種により惹起される細胞死をLTRPC2が仲介することが示唆された。活性酸素種はTRP関連チャネルの正常な活性維持にも深く関与しており、細胞レドックス状態を感受してCaシグナルに変換するTRPの生理学的意義が示唆された。
