延髄孤束核での一酸化窒素合成酵素の単一分子の動きの観察

研究概要

延髄孤束核(nucleus tractus solitarius, NTS)で一酸化窒素合成酵素(NO synthase, NOS)がどれくらいの頻度で合成されどれぐらいの時間で消滅していくかを調べるため、NTS内へ神経型NOS(neuronal NOS, nNOS)の遺伝子と、nNOSの遺伝子の下流にgreen fluorescent protein(GFP,染色しなくともそれ自身が蛍光で光る蛋白質)の遺伝子を組み込んだベクターを導入することを目指し、以下の実験を行った。
1.先ず、in vivoで脳へプラスミドベクターを用い遺伝子導入できる簡便な技術が開発されていない。そこで、nN0Sの遺伝子を組み込んでいないGFP Fusion TOPO Cloning Vectorsを作りトランスフェクション試薬とともにラットの脳へマイクロインジェクションし、その後2日目、3日目、5日目、7日目、10日目に脳(各々n=5)を取り出し、蛍光を観察した。
2.その結果、5日目では5匹中4匹で蛍光が観察された。蛍光強度は、5日目が番上昇していた。遺伝子発現は5日目が番強いと考えられた。
3.次に、β-galを組み込んだ遺伝子を脳へ注入した。しかし、この場合はβ-galで染色されず、少し大き目の遺伝子では従来のトランスフェクション試薬では脳への導入効率が悪いと思われた。
4.トランスフェクション試薬として、HVJ-envelope法が最近開発され、その方法を使い現在検討中である。
5.また、nNOSの遺伝子をGFP Fusion TOPO Cloning Vectors内へ組み込む実験を始めている。
