新規アポトーシス調節蛋白貭ASCの機能解析

研究概要	ASCはcaspase-recruitment domain(CARD)とPYRIN domain(PYD)という2つの結合ドメインを持つアダプター分子であることを明らかにしました。私達のこの結論は、「PYDはDD,DED,CARDが属するdeath domain familyの第4番目のメンバーである。」とする米国のグループの結論によっても支持されました。ASCはアポトーシス刺激によって凝集する分子として同定したものですが、この凝集はPYDまたはCARDを介する結合によって生じたものだと解釈することができます。我々の報告以降、PYDモチーフを持つ分子がバイオインフォマティクスによって次々に発見され、現在ではヒトだけでも15種類近くのPYD分子の存在が報告されています。アポトーシス及びそれに関連するNF-kBの活性化は分子集合による「近接活性化」が引き金になって起ることが知られています。ですから、次の課題は「ASCの多量体形成のトリガーは何か?」、「どのようなエフェクター分子がこの多量体形成で近接活性化されるか?」ということを考えなければなりません。ヒトの正常組織では、ASCは未熟な血球前駆細胞や皮膚の基底細胞では発現しておらず、成熟した末梢血の単球・顆粒球系細胞や皮膚の角化細胞で高い発現が観察されます。また、虫垂炎や劇症肝炎など、炎症部位でASCの発現が著明に増加します。この現象は、炎症反応におけるASCの重要性を示すとともに、量的な変化が多量体形成の一つのファクターである可能性を示唆しています。最近、家族性地中海熱および家族性非発熱性蕁麻疹の原因遺伝子産物であるpyrinとcryopyrin/PYPAF-1がASCと結合し、細胞死またはNF-kBを活性化することが明らかにされました。この結果は炎症疾患の分子機構解明にはASCの機能解析が欠かせないことを示しています。
