酸性スフィンゴ糖脂質による神経系の維持と修復の分子機構

研究概要	主に、複合型ガングリオシドを欠損するGM2/GD2合成酵素遺伝子のノックアウト(KO)マウスを用いて、ガングリオシド糖鎖が神経系の維持と修復において果たす役割について検討を行った。GM2/GD2合成酵素遺伝子KOマウスの行動異常の検討として、各週齡のオス/メスマウスの歩行をフットプリントにて検討したところ、オスでは35週より前肢の引きずり歩行が始まり、50週で後肢も引きずるようになった。メスでは70週で前肢、80週で後肢の異常を認めた。また、ロータロッド試験では、オス・メスともに既に12週齡から著明な異常を認めた。しかし、繰り返し試行するとメスでは一定の改善が見られるのに比して、オスでは改善が見られなかった。以上より、ガングリオシド欠失に基づく神経変性が脊髄では上位から下方に拡大すること、またその感受性において雌雄の間に大きな相違があることが判明した。電顕による観察では、脊髄後根神経節および脊髄後角のニューロンの変性/死やグリア細胞の肥大化が認められ、ガングリオシドが神経組織の維持に必須であることが示された。さらにグリアの突起が特に血管周囲に増生していたため、グリア機能の一つとして血液・脳関門(BBB)機能をHRP注入にて検討したところ、4週齡のKOマウスにおいて血管外へのHRP漏出が見られたことから、KOマウスでは早期よりBBB機能に異常があることが示唆された。現在、KOマウスと野生型マウスの脊髄よりRNAを抽出してサブトラクションを行い、変性に関与する遺伝子、変性を代償する遺伝子の同定を行っている。また、舌下神経切断後の再生能に関して、KOマウスでは著明な低下があり、外来性ガングリオシドの投与にてかなりの改善が得られることが判明しているが、現在、有効な効果を示す糖脂質糖鎖の特異性と、その機序の解明のために、再生過程における種々のシグナル分子や神経栄養因子(受容体)の発現を検討中である。
