新奇な転写活性化機構を構成するプロモーター上のステム・ループ構造

研究概要	腺組織に特異的に発現する糖転移酵素Gal NAc T3の遺伝子プロモーター領域や抗癌剤(シスプラチン)耐性細胞で発現亢進するインターロイキン13受容体の遺伝子プロモーター領域は、ともにG・C richな配列であり、約60塩基からなるステム・ループ構造をin vivoで形成し、転写制御に重要な役割を果たしていることが示唆されている。また、0.5Kbという極めて近接して配置されている赤血球型ピルビン酸キナーゼ遺伝子プロモーター領域と肝型ピルピン酸キナーゼ遺伝子プロモーター領域においても、両プロモーターの中間部にはin vivoでステム・ループ構造が形成されていることが、一本鎖状態にあるチミン塩基に対して反応性をもつ過マンガン酸カリウムを用いた解析から明らかになった。ヌクレオソーム構造のリンカー部位を優先的に切断するマイクロコッカル・ヌクレアーゼを用いて、ステム・ループ構造領域の周辺のクロマチン構造をゲノム塩基配列レベルで解析した結果、上記のステム・ループ構造はクロマチン構造遮蔽素子として機能している可能性が浮上してきた。これらのステム・ループ構造領域は、当初想定していた約60塩基の範囲にとどまらず、同様なステム・ループが林立している模様であり、ステム・ループ構造をとっている領域の厳密な絞り込みが急務である。さらに、そうした領域的な絞り込み作業に続き、クロマチン構造遮蔽素子としての機能検証を早急に進める必要があると考えている。
