DNAマイクロアレイとマイクロ・ダイゼクションを併用した癌の遺伝子解析

研究概要	【目的】レーザー顕微鏡下マイクロダイセクション(Laser micordissection ; LMD)とDNAマイクロアレイを用いて消化器癌に関する重要な遺伝子の発現プロファイルを外科切除標本を用いて検索した。次に各症例の術前生検標本についても同様に調べ、本法の術前診断への臨床応用を試みた。【方法】胃癌5例の癌部、非癌部、および転移巣(リンパ節)より総計15検体採取した。各検体20枚ずつ(合計300枚)の凍結切片を作成し、LMDを用いて目的の細胞のみを一検体につき百から1万個採取した。全RNAよりT7-based RNA法を用いてmRNAのみを増幅し、425種類の癌関連遺伝子搭載のHuman Cancer Chip ver2.1(宝酒造)を用いて、(1)癌部-非癌部間または(2)原発巣-転移巣間で発現差のある遺伝子群を同定し、発現プロファイルを得た。【結果】(1)癌部と非癌部の比較;癌部において過剰発現していた遺伝子は細胞周期関連遺伝子としてRB protein、MAP kinase、neutrophic tyrosine kinase、増殖因子としてInsulin like growth factor 2、Platelet derived growth factor (PDGF)等であった。(2)転移巣と原発巣の比較:転移巣において過剰発現していた遺伝子は細胞周期関連遺伝子としてtyrosine kinase 2、蛋白分解酵素関連遺伝子としてcollagen type III-α、増殖因子としてPDGF、転写因子としてEukaryotic translation initiation factor-2、接着因子としてCadherin等が認められた。これらの既知の遺伝子およびESTについては発現差を確認した。【結論】LMDおよびDNAマイクロアレイを用いたことから、非目的細胞の混入無く癌化あるいは転移において重要な遺伝子群を精確に同定し得た。LMDおよびDNAマイクロアレイを用いた生検標本の評価は癌の悪性度あるいは転移の予測をする上で今後極めて重要になると考えられた。
