HTLVおよびHHV産生細胞の各種抗原発現に関する免疫電子顕微鏡的研究

研究概要	HTLV-IとHTLV-II産生細胞株におけるT細胞受容体(TCR)とそのα、βサブユニット発現に関しては、共通点が認められており、いずれも細胞株となった時点でαサブユニットの消失に伴い、βサブユニットは年余に亘り産生されるにも拘わらず、TCRの細胞表面への発現は微弱ないし消失することが明らかとなった。また、CD4、CD8発現も減弱ないし消失しており、CD3についても同様であった。これらTCRやCD3、CD4などの発現異常が腫瘍細胞のT細胞としての機能異常に深い関係を有していると考えられる。また、レクチンとの結合性においてはフェリチン抗体直接法によりRCA-I、WGA、ConAと結合反応が弱くなり、UEA-Iでは全く標識されなかった。 Human herpes virus(HHV-8)に関しては、TPA処理によりその微細構造を明らかにすることができ、他のヘルペスウイルス〔HHV-4(EBV)、HHV-5(CMV)、HHV-6、HHV-7〕とは形態的に異なることを明らかにした。また、外径260nmにもおよぶmegalovirusの存在を明らかにした。目下、HHV-6が形態的に観察しにくい点を考慮して、HTLV-I産生細胞(MT-4)との同時感染により、HHV-6を顕在化させる試みを実施中である。
