赤痢菌の上皮細胞間感染におけるアクチン重合機構の研究

研究概要

赤痢菌の細胞内運動の原動力となるアクチン重合機構には、菌の大プラスミド上にコードされているVirG蛋白が必要である。これまでにVirG蛋白の機能領域を解析し、さらに本蛋白と結合する宿主側因子としてビンキュリンおよびNeural-Wiskott Aldrich syndrome protein (N-WASP)を同定した。しかし、感染細胞内においてVirGとN-WASPがどのような相互作用を生じ、これにより細胞内のどのような因子が動員されてアクチンコメット形成に至るのか、さらにアクチン重合によって運動能を獲得した赤痢菌がどのように隣接細胞へ再侵入を行うのか、それらの分子機構は未だ明らかではない。そこで本研究では、VirGによるアクチン重合機構においてN-WASPに結合するシグナル伝達分子やアクチン重合制御因子を同定することを企図した。
平成13年度:赤痢菌感染細胞内における上記シグナル伝達分子およびアクチン重合制御因子の時間的・空間的動態とその役割の解析
哺乳動物細胞においてWASPファミリーのうちWASPは血球系細胞にのみ発現するが、N-WASPやWAVEは普遍的に発現すると考えられている。しかしマクロファージ、多形核白血球といった血球系細胞にはN-WASPがほとんど発現していないことを見い出した。これらの細胞ではWASPが大量に発現していた。VirGはN-WASPとのみ特異的に結合することから、赤痢菌が感染したマクロファージの細胞質内ではVirGと結合するN-WASPがほとんど存在しないがためにアクチン重合が生じず、その結果アクチンコメットの形成がほとんど起こらないことが明らかになった。このことはVirG-N-WASPの特異的結合が赤痢菌の細胞内運動における宿主細胞特異性を決定づけているものと考えられる。
