インフルエンザ菌の多剤耐性における排出蛋白の役割

研究概要	インフルエンザ菌の抗菌薬に対する耐性機構には、β-ラクタム薬の標的部位であるペニシリン結合蛋自質(PBP)が変異した耐性菌BLNAR(β-lactamase-negative ampicillin-resistant H.influenzae)や排出ポンプによる耐性化などが知られている。私どもは、インフルエンザ菌(H.influenzae)のRND型AcrAB異物排出システムが、Macrolide系抗生物質をはじめいくつかの抗菌性化合物の排出に寄与している事を明らかにしてきた。本年度私どもはAcrAB異物排出システムのさらなる機能の解明を目的に特異抗体の作製、及び特異抗体を用いてAcrAB排出システムの発現量とMacrolide系抗生物質耐性との相関性について検討を行った。方法としてはまずデータベースを用いて推定された排出システム遺伝子をPCR法により増幅し、His-tagを含む発現ベクターにクローン化した。次に発現ベクターを誘導しタンパクを過剰発現させた後、Western Immunoblot法により特異的タンパク質を検出した。このタンパク質をカラムを用いて精製した後、濃縮し抗体作製の抗原タンパク質として使用した。マウスポリクローナル抗体の作製はタカラ・バイオ研究所に依頼した。この作製された抗体を用いてATCC10211及び臨床分離株(48株)におけるAcrAの発現量を定量し、さらに感受性を寒天平板希釈法により求め、感受性と発現量との関連性を比較検討した。今回作製した抗体をWestern Immunoblot法でATCC10211及び臨床分離株(48株)においてAcrAの発現を確認したところすべての株においてAcrAの推定分子量と思われる位置に発現を確認できた。この事よりこのインフルエンザ菌のAcrAB異物排出システムは構成的に発現している事が確認できた。臨床分離株(48株)中にAcrA過剰発現株が検出され、その株においてMacrolide系抗生物質に対する感受性の低下が見られた事によりAcrAB異物排出システムがMacrolide系抗生物質耐性の一因となっている可能性が示唆された。
