EBウイルスLMP1による上皮細胞癌化・悪性化機構の解析

研究概要	EBウイルスの癌遺伝子のひとつであるLatent Membrane Protein-1(LMP1)の上咽頭癌組織での発現は浸潤性・悪性度などとよく相関することをこれまでに報告してきた。本研究ではLMP1による上皮細胞癌化のモデルとして腎上皮細胞由来MDCK細胞をLMP1でトランスフォームすることに成功した。トランスフォーム細胞は高い浸潤活性を示した。浸潤性・悪性度に関わる関連遺伝子を検索した結果LMP1の発現は転写因子であるEts-1の発現を誘導し、その結果として細胞の形態変化、細胞運動能の亢進が起こることを明らかにすると共にその分子機構を解析した(Oncogene 30、1764-1771、2000)。さらにEts-1のDominant Negative変異体を発現することにより細胞運動能の亢進を抑制できることを示した。次にEts-1によって転写制御を受ける可能性のある遺伝子について上咽頭癌組織における発現を検討した。とりわけLMP1の発現は血管新生ともよく相関することを見い出したので血管新生関連遺伝子を中心に検討を行い、Interoleukin-8(IL8)の発現がLMP1発現と相関することを見い出し、in vitroの実験でもLMP1がIL8の発現を誘導することを示した(Clinical Cancer Res., 7, 1946-1951,2001)。一方、上皮細胞の運動性亢進という観点からHepatocyte Growth Factor (HGF) のリガンドであるc-Metに着目し、上咽頭癌組織ではLMP1とc-Metの発現が相関すること、in vitroの実験でLMP1がc-Metの発現を誘導することを見い出した(American Journal of Pathology, 159, 27-33)。以上、本研究によりLMP1は上皮細胞の癌化のみならず悪性化にも関与することをin vivoおよびin vitroの実験により明らかにした。
