GC/MS/MSによるヒト体液中乱用薬物の高感度分析法の確立及びマニュアル化

研究概要

本年度は、昨年に引き続いて、電子衝撃(El)法と化学イオン化(Cl)法との比較検討を続行した。El法は再現性が良好であるものの、十分な擬分子イオンが得られず、一方でCl法は再現性に問題があったが、得られたペチジンのピークのスペクトルは殆どが[M+1]^+イオンであり、最終的に、我々は親イオンを分離するためのイオン化としてはClイオン化法を採用した。内部標準として、当初は1-フェニルピペリジンを用いたが、固相抽出の際にカラムに吸着しないため、ペチジンに構造の類似した4'-ピペリジノアセトフェノン(4'-PAP)を採用し、BondElut C_18を用いた固相抽出によって体液中から精製した。MS/MS条件は以下の通り。ペチジンは、親イオン(m/z248)をレゾナントモードで分解し、m/z174及び202の娘イオンをスキャンした。内部標準は親イオン(m/z204)をノンレゾナントモードで分解し、m/z162の娘イオンをスキャンし、両ピークの面積比から濃度を推定した。両薬物の回収率は、89%から109%であった。また、1.25から40ng/mlの範囲で直線性を検討したが、比較的高濃度の領域で直線から外れる傾向が認められた。この原因については、トラップ内の微細な環境の違いが、イオン化効率に大きく影響している可能性が示唆された。これを回避するためには、重水素で標識した内部標準を用いることが必要である。現在、データの整理を行っており、専門誌に投稿する予定である。
今後は、同じく麻薬性鎮痛剤のペンタゾシンについてもイオン化条件等の検討を行い、高感度の検出法を確立する予定である。
