虐待児童とストレス負荷動物の胸腺退縮に関わるチロシンリン酸化情報伝達経路の解析

研究概要

ストレス負荷した動物モデルと実際の虐待児童の退縮胸腺におけるチロシンリン酸化反応の程度をコントロールのものと比較検討した。コントロール胸腺では、チロシンリン酸化蛋白質は髄質のハッサル小体に豊富に発現していた。また、ハッサル小体にはサークファミリーに属するチロシンリン酸化酵素及びJAKチロシンリン酸化酵素が局在していることが免疫組織の結果から明らかとなった。ハッサル小体が胸腺髄質の上皮細胞の分化したものであることから、これらの結東はチロシンリン酸化反応が胸腺髄質の上皮細胞の分化に役割を果たしていることを示唆している。一方、退縮胸腺では、チロシンリン酸化蛋白質の発現が低下すると共にハッサル小体の縮小が生じていることが明らかとなった。退縮胸腺ではチロシンリン酸化反応が負に調節されており、これが胸腺髄質の上皮細胞の分化に影響を与え、ハッサル小体の形態変化をもたらしていると考えられる。ハッサル小体に局在していることが明らかとなったJAKと呼ばれるチロシンリン酸化酵素は、サイトカインの情報伝達経路に重要な役割を果たしていることがすでに報告されている。我々が、今回研究を行ったのは細胞内のチロシンリン酸化経路であり、一般に細胞内の情報伝達経路は細胞外からもたらされるサイトカインなどの情報伝達物質によりその活性が調節されている。従って胸腺が退縮をきたすようなストレス状態では生体中のサイトカインの量が減少しており、この結果細胞内で特にJAKを介するチロシンリン酸化反応が抑制されハッサル小体の形態変化がもたらされることが考えられる。サイトカインは血中で測定可能であることから、本研究は虐待児童の診断にサイトカインの測定が有効である可能性を示したものといえる。実際の虐待児童の血中のサイトカインの定量、またどういった種類のサイトカインが実際に減少しているのかなどについて研究をすすめる必要がある。
