ヒト単球遊走因子(hMCP-1)転写制御解析-hMcp-1発現抑制によるリウマチ関節炎遺伝子治療の試み

研究概要	MCP-1の転写にNF-κBとSp1が重要である。しかしながらMCP-1の発現をすべて転写レベルで説明することはできない。Bリンパ球は本来NF-κBがconstitutiveに活性化されていることが判明しているが、我々が末梢血から精製したBリンパ球にはその発現を認めなかった。興味深いことに、このBリンパ球をIL-10とanti-CD40の存在下で5日間培養したところ、Bリンパ球の分化に伴いIgが発現し、さらにMCP-1の発現も確認された。この発現はLPSおよびTPA刺激により増強したが、この過程において一貫してNF-κBの活性化に変化はなかった。多くのBリンパ球由来のcell lineをスクリーニングした結果RPMI8226細胞で分化したBリンパ球と同様にhMCP-1の発現を確認し得た。この細胞をTPAで刺激するとhMCP-1の発現は極めて強くなることも確認されたが、EMSAにおけるNF-κBの活性化は確認できなかった。RPMI8226細胞におけるhMCP-1mRNAの発現亢進の機序は転写に非依存性であることが想像された。そこで次に転写後のmRNAのstabilityに関する解析を行った。RPMI8226細胞をTPA及びLPSで4時間刺激した後、アクチノマイシンDでmRNAの合成を止めノーザンプロットでその半減期を調べた。刺激のない状態で半減期は約3時間、刺激後は約16時間へと延長していた。MRNAの安定性には3'側のAU-richなエレメントが重要な働きをすることが知られている。Human, mouse、等MCP-1ホモログの3'領域を比較したところAU-richな領域があることが判明した。過去の研究も含めMCP-1の発現を制御する因子がおおよそ明らかになったことから、我々は関節炎におけるMCP-1の発現異常の機序、さらにMCP-1過剰発現が病態形成にどの程度関与しているか明らかにするためIn vivoの実験を開始した。vivoへの遺伝子導入法としてアデノウイルスを選択した。マウスMCP-1cDNAを発現カセットに有するリコンビナントアデノウイルスを作成した。ウイルスはCsClの不連続クッションで遠心精製した後、適当なMOIで培養細胞に感染させmouse MCP-1の発現を確認した。
