膵癌細胞におけるビタミンD,TGFβシグナル伝達系クロストークの検討

研究概要	膵癌細胞を用いてビタミンD,TGFβシグナル伝達系クロストークの検討を行った。ビタミンD3(vitD)、TGFβ反応性細胞SUP-1においてTGFβシグナル伝達系の終末に位置するsmad4(PC4)のNorthern blottingを行うと、発現を認めなかったことより、vitDとTGFβシグナル伝達系にクロストークの存在が考えられた。SUP-1にビタミンDレセプターのresponse elementであるVDRE3-Lucをtransfectionして、vitDとTGFβを反応させVDRの転写活性能を検討したが、vitDにて転写能の亢進を認めたが、TGFβでは認めなかった。従って、今回の検討では両シグナル伝達系のクロストークの証明はできなかった。膵癌との鑑別が問題となる自己免疫性膵炎について検討をおこなった。本疾患ではDRB10405-DQB10401 haplotypeの頻度が有為に高く、これらclass II抗原でpresentationされる特定の抗原ペプチドに対する免疫応答が発症の引き金になっているのではないかと考えられた。自己免疫性膵炎の患者血清に特異的に反応する、25kDaの蛋白質を検出した。この蛋白質は、carbonic anhydrase(CA)とは異なり、等電点は7.5付近で、極めて不溶性であることより、支持組織等に存在する不溶性の細胞間質・繊維等の構成成分である可能性が高いと思われる。
