肝内樹状細胞の単離とその機能の特性に関する研究

研究概要	生体内で最も強力な抗原提示細胞である樹状細胞(DC)がウイルス性肝炎や自己免疫性疾患および悪性腫瘍で機能異常があることを報告してきた。しかし、これらは、脾臓や末梢血のDCについての検討であり、実際の病変の現場である肝内のDCについては検討されていない。今回、C57BL/6マウス肝からDCの単離を行った。成熟したDCである脾DCと比較して次のような特徴がみられた。1.フローサイトメトリーで細胞表面マーカーの発現を解析すると、肝DCはMHC class II,CD86,CD80が低発現であった。2.FITC標識dextranを用いた抗原の取り込み能の検討では、強い抗原取り込み能がみられた。3.異系同種T細胞と混合培養すると、T細胞幼弱化補助能が極めて弱かった。4.培養上清中のサイトカイン産生をELISAで測定すると、IL-12の産生は極めて低く、IL-10の産生がみられた。次に、特異抗原としてHBs抗原、Keyhole limpet hemocyanin(KLH)を用いて抗原特異的な免疫応答について検討した。1.肝DCに抗原(HBsAg,KLH)をパルスするとMHC class II,CD86の発現が増強し、異系同種T細胞幼弱化補助能も上昇した。 2.HBsAg,KLH感作T細胞と抗原をパルスした肝DCを混合培養すると、抗原特異的T細胞の幼弱化の誘導が可能であった。また、培養上清中には多量ののIL-12,IFN-γの産生がみられた。以上から、1.肝DCは未成熟なDCであること、2.肝DCは抗原を取り込んだ後には抗原特異的な免疫応答が誘導できることが明らかとなった。
