脳内の内因性神経ペプチドが肝・胆・膵の生理機能および病態に及ぼす作用に関する研究

研究概要	脳内の内因性神経ペプチド(Thyrotropin-releasing hormone : TRH)が肝血流に影響を及ぼすかどうかをラットを使って検討した。ウィスター系雄性ラットをウレタン麻酔下で開腹し、水素ガスクリアランス法およびレーザードップラー法にて肝血流量を測定した。延髄迷走神経運動核でのTRH放出を刺激することが知られているカイニン酸(0.5〜20pg)を縫線核にmicroinjectionして肝血流量の変化を120分間観察した。またカイニン酸投与の5分前にpolyclonal TRH抗体を脳槽内に投与してカイニン酸刺激による肝血流変化に対する効果も検討した。その結果、カイニン酸の縫線核へのmicroinjectionにより、投与後30分をピークとする肝血流の増加が用量依存性に観察された。本作用はTRH抗体の脳槽内前投与および迷走神経切断術によって消失した。本結果より、カイニン酸のmicroinjectionによる縫線核への刺激が脳内の内因性TRH放出を促して迷走神経経由で肝血流を増加させることが確認され、脳内の内因性TRHが生理作用として肝血流を制御していることが示唆された。
