抗消化管ムチン単クローン抗体の系統的開発とその病態生理学的研究への応用

研究概要

申請段階で掲げた4項目の目標毎に研究の伸展の経過を報告する。
1)糖鎖構造情報解析の手段としての単クローン抗体の確立と供給:この研究の為に必要となるエピトープ糖鎖を含むムチンの分離精製を行った。本年度は、ラット胃ムチン(RGM21,RGM26)、ラット小腸ムチン(HCM31)、ヒト胃液ムチン(HGM75)に対象をしぼり、出来る限り多くのムチンを集める努力を継続した。次年度はこのうち、RGM21、HCM31およびHGM75に焦点を絞って、これらの抗体のエピトープ糖鎖を分離精製する試みを行う予定である。
2)Helicobacter pylori(Hp)感染に対するムチンの関与の検討:ヒト切除胃よりムチンを精製し、表面プラズモン共鳴装置(IAsys)を用いた、Hpとムチンとの結合を測定する実験系を開発した。この方法によって、胃体部と幽門部から得たムチンのHpとの結合性の違いが測定できた。次年度は、ヒト胃ムチンをさらに精製し、Hpと特異的に結合するムチンの性質を明らかにする予定である。
3)実験潰瘍の治癒期における胃粘膜め免疫組織化学的検討:ラット酢酸潰瘍の治癒過程の胃粘膜から組織切片を作成し、これまでに開発した単クローン抗体による免疫組織化学染色を行った。その結果、従来より指摘されている、再生上皮に幽門腺型の粘液の発現がみられる(Biomed Res,2000)という知見に加えて、正常の胃体部・幽門部には存在しないが、HCM31で検出できる型のシアロムチンが粘膜再生の比較的早期(15-50日)に一過性に出現することがわかった(Scand J Gastroent,2001)。次年度はHCM31の抗原となるムチンの発現の意義をさらに追求する予定である。
4)病理組織標本を用いた免疫組織化学染色による各単クローン抗体の有用性の検討:共同研究を展開している岡安(本学医学部病理)らのグループによってHGM75およびHGM72の有用性が確認された(Histopathol,2000)。
